Nexium® / PPI Consumer Warning

The U.S. Food and Drug Administration has issued a consumer warning regarding a possible increased risk of fractures with high doses or long-term use of the heartburn medication Nexium®.

Possible injuries include:

  • Hip factures
  • Spine fractures
  • Wrist fractures
  • Bone deterioration
  • Other injuries

Nexium, manufactured by AstraZeneca, is a proton pump inhibitor (PPI) used to treat conditions such as gastroesophageal reflux disease (heartburn), stomach and small intestine ulcers, and inflammation of the esophagus.

A British study published in the December 2006 issue of the Journal of the American Medical Association found prolonged and/or heavy use of Nexium increases the risk of hip fractures in individuals over 50 years old. Nexium works by blocking the enzyme in the stomach wall that produces stomach acid. The reduction of stomach acid can make it harder for the body to absorb some nutrients, including calcium, which can lead to weakened bones and fractures.

Other PPIs included in this consumer warning are dexlansoprazole (Dexilant®), omeprazole (Prilosec®, Zegerid®), lansoprazole (Prevacid®), pantoprazole (Protonix®), and rabeprazole (Aciphex®).

“Epidemiology studies suggest a possible increased risk of bone fractures with the use of proton pump inhibitors for one year or longer, or at high doses,” said Joyce Korvick, M.D., deputy director for safety in FDA’s Division of Gastroenterology Products. “Because these products are used by a great number of people, it’s important for the public to be aware of this possible increased risk and, when prescribing proton pump inhibitors, health care professionals should consider whether a lower dose or shorter duration of therapy would adequately treat the patient’s condition.”
